=== 5α-Reductase inhibition === Progesterone is a substrate for 5α-reductase, and has been found to act as a competitive inhibitor of this enzyme in vitro in a variety of studies. In one study, it showed IC50Tooltip half-maximal inhibitory concentration values of 1,375 nM and 88 nM (in the presence of 50 nM androstenedione as the substrate) for 5α-reductase types 1 and 2, respectively. 5α-Reductase is highly expressed in skin, hair follicles, and prostate gland, and is responsible for the transformation of testosterone into the several-fold more potent androgen DHT in such tissues. As such, it has been suggested that progesterone might possess some antiandrogenic effect via acting as a 5α-reductase inhibitor. However, inhibition of 5α-reductase by progesterone is described as a weak effect that has only been demonstrated in vitro and at supraphysiological concentrations. In accordance, physiological levels of circulating progesterone have not been found to importantly influence circulating DHT concentrations. Congenital 5α-reductase 2 deficiency is a rare intersex condition which is associated with ambiguous genitalia in male fetuses due to a deficiency in DHT production during genital differentiation. Experimental prenatal exposure to established 5α-reductase inhibitors like finasteride has been found to produce similar feminized genital defects in male animals including rodents and monkeys. In contrast, exogenous administration of progesterone to pregnant rodents and monkeys has resulted in minimal abnormality in either male or female pups.
== Contraindications == Suvorexant is contraindicated in people with narcolepsy as it may worsen their symptoms. This is its only absolute contraindication. Suvorexant has not been studied in people with severe hepatic impairment and is not recommended in these individuals due to the likelihood of increased suvorexant exposure. On the other hand, suvorexant may be used in people with mild-to-moderate hepatic impairment as well as renal impairment of any severity and no dose adjustment is necessary in these situations. Concomitant use of suvorexant with strong CYP3A4 inhibitors is not recommended due to potential for increased suvorexant exposure while concomitant use of suvorexant with strong CYP3A4 inducers may result in loss of suvorexant exposure and effectiveness. Suvorexant should be used carefully in people with a history of drug misuse or alcoholism due to its drug-liking effects and possible misuse potential at doses higher than those approved for therapeutic use. Similarly, suvorexant should be used carefully in people with a history of depression or suicidality as it may rarely increase suicidal ideation. The medication is indicated for use in adults and the elderly but has not been studied in children and adolescents and hence is not recommended for these individuals. Suvorexant has shown teratogenic effects in animals such as decreased body weight at doses much higher than the equivalents of those approved for therapeutic use in humans.

=== Use of hybrid warfare === The Trump administration's actions against Greenland and Denmark, initially referred to as the Greenland crisis, have been described by academics and commentators as hybrid warfare, a broad umbrella concept that may include the use of political warfare, fake news, diplomacy, lawfare, regime change, and foreign electoral intervention, among other methods. Greenland was united in condemnation of the US, describing it as "an entirely new and unsettling situation", given that Greenland lacked realistic countermeasures. Danish media interviewed several experts who said that the US actions against Greenland clearly amount to hybrid warfare. André Ken Jakobsson, a researcher on hybrid warfare, said, already in the spring of 2025, that "the tools the United States uses to achieve its objectives [in Greenland] also fall within the gray zone typically associated with hybrid warfare". Jakobsson believes the United States is now deliberately seeking to fracture the Kingdom of Denmark with the use of intelligence agencies and that the US has been using all of its means short of conventional war in its campaign to try to get Greenland. Speaking on US hybrid warfare and espionage, DR commentator Niels Fastrup said that the US has shifted from an attempt to buy Greenland in 2019 to hybrid influence operations, conducted by private individuals close to Trump.

=== Xen and final release (2015–2020) === The release of the Xen part of the game had been the most difficult, since the team wanted to redesign the levels to overcome the poor perception that they had in Half-Life's original release. The team said, "We want our version of Xen to feel like it really belongs with the rest of the game in terms of mechanics, cohesion and progression," while at the same time, they wanted "to push the boundaries and explore this unique and varied setting; to build an experience that feels both fresh and familiar to players from all walks of Half-Life veterancy." Developing their new version of Xen was a chicken-or-the-egg dilemma, as without level design it was difficult to develop art assets, and without art assets it was hard to come up with cohesive level designs. They also wanted to give more story elements there, such as why human scientists were studying the world of Xen in the first place, trying to capture the same type of world-building by level design that Valve had been able to with the first parts of Half-Life. They also significantly reworked the boss battles to be more challenging and representative of the area they had in mind. Ultimately, the team expanded out Xen from about a one-hour experience in the original Half-Life to four hours in Black Mesa.
== Interactions == Zopiclone also interacts with trimipramine and caffeine. Alcohol has an additive effect when combined with zopiclone, enhancing the adverse effects including the overdose potential of zopiclone significantly. Due to these risks and the increased risk for dependence, alcohol should be avoided when using zopiclone. Erythromycin appears to increase the absorption rate of zopiclone and prolong its elimination half-life, leading to increased plasma levels and more pronounced effects. Itraconazole has a similar effect on zopiclone pharmacokinetics as erythromycin. The elderly may be particularly sensitive to the erythromycin and itraconazole drug interaction with zopiclone. Temporary dosage reduction during combined therapy may be required, especially in the elderly. Rifampicin causes a very notable reduction in half-life of zopiclone and peak plasma levels, which results in a large reduction in the hypnotic effect of zopiclone. Phenytoin and carbamazepine may also provoke similar interactions. Ketoconazole and sulfaphenazole interfere with the metabolism of zopiclone. Nefazodone impairs the metabolism of zopiclone leading to increased zopiclone levels and marked next-day sedation.
