Stability depends on pH, temperature, oxygen exposure, and trace metals. Aqueous solutions of reduced glutathione are susceptible to oxidation, especially when neutral or alkaline and exposed to air. Transition metal ions can catalyze thiol oxidation, so chelators and inert atmospheres are sometimes used in research settings. Standards are typically stored cold and desiccated, with limited freeze-thaw cycles. Questions remain about how closely in vitro stability data reflect the behavior of glutathione within intact cells and tissues.
Measuring glutathione requires attention to oxidation during sample handling, because GSH in biological samples can convert to GSSG or form mixed disulfides with proteins after collection. Acidic extraction, rapid cooling, and chelating agents are commonly used to limit such changes. Analytical methods usually distinguish free reduced glutathione, total glutathione, and protein-bound forms. Because these forms have different stability and reactivity, reported values depend heavily on the preparation protocol. No single preparation is universally suitable for every biological matrix or analytical goal.
Biosynthesis proceeds in two ATP-dependent steps. First, glutamate-cysteine ligase joins glutamate and cysteine. Second, glutathione synthetase adds glycine to the intermediate. The pathway is regulated by cysteine availability, enzyme expression, and feedback inhibition by glutathione itself. Liver tissue has a particularly high capacity for synthesis and export. Because the molecule is made inside cells, circulating glutathione reflects a balance of release, uptake, and breakdown rather than simple dietary supply.
Functionally, glutathione supports redox balance by donating electrons and becoming oxidized. It also serves as a cofactor for enzymes such as glutathione peroxidases and glutathione S-transferases. These enzymes participate in peroxide reduction and in conjugation reactions that help process reactive molecules. Separate from antioxidant roles, glutathione can modify protein cysteines through S-glutathionylation, influencing enzyme activity and signaling. Research continues to examine how these chemical roles translate into whole-organism effects.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. Its glutamate-cysteine linkage uses the gamma-carboxyl group of glutamate, a feature that resists standard peptidases. The cysteine residue provides a thiol group, which gives the molecule its reducing character. In cells, glutathione is often the most abundant small-molecule thiol, with concentrations varying widely by tissue and compartment. It exists mainly in a reduced form called GSH, while oxidation produces a disulfide-linked dimer called GSSG.
| Property | Value | Notes |
|---|---|---|
| Reduced form | GSH | Main intracellular thiol |
| Oxidized form | GSSG | Disulfide dimer of two GSH molecules |
| Common separation method | Reversed-phase HPLC | Often with ion-pairing or derivatization |
| Typical detection | Fluorescence or mass spectrometry | UV detection is also used in some assays |
| Storage of standards | -20 °C or below, desiccated | Limit freeze-thaw and moisture exposure |
Quantification of glutathione in biological or food samples commonly uses liquid chromatography coupled to ultraviolet, fluorescence, electrochemical, or mass spectrometric detection. Because the thiol group oxidizes readily, samples are often acidified or derivatized immediately after collection to stabilize reduced glutathione. Enzymatic recycling assays and colorimetric kits offer higher throughput but generally lower specificity than chromatographic methods. Mass spectrometry can distinguish glutathione from related thiols and allow simultaneous measurement of oxidized forms. Reported concentrations depend strongly on sample type, extraction procedure, and analytical platform.
Glutathione reference materials are sensitive to oxygen, light, and elevated temperature. Solid material is typically stored desiccated at -20 °C or below, while solutions require tighter control because thiol oxidation proceeds faster in liquid form. Aqueous solutions are often prepared fresh, kept cold, and protected from air; some protocols add acid or chelating agents to slow metal-catalyzed oxidation. Repeated freeze-thaw cycles can accelerate degradation and should be avoided. Stability data vary by matrix, so laboratories usually verify performance with their own storage conditions.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. It occurs in nearly all living cells, with highest concentrations in liver, kidney, and red blood cells, and exists in reduced (GSH) and oxidized disulfide (GSSG) forms. The cysteine thiol group enables reversible oxidation and reduction reactions. This property makes glutathione a central participant in cellular redox balance. The balance between these forms is often used as an indicator of oxidative stress.
Glutathione synthesis proceeds in two ATP-dependent steps catalyzed by glutamate-cysteine ligase and glutathione synthetase. The first step joins glutamate and cysteine to form gamma-glutamylcysteine and is generally rate-limiting. The second step adds glycine to complete the tripeptide. Cysteine availability, feedback inhibition by glutathione, and oxidative conditions influence flux through this pathway. The pathway is conserved across many organisms, and degradation by gamma-glutamyl transpeptidase and related peptidases recycles amino acids for new synthesis.
Within cells, glutathione serves as a cofactor for glutathione peroxidases and glutathione S-transferases. These enzymes reduce hydrogen peroxide and organic peroxides or conjugate electrophilic compounds to the thiol group. The resulting conjugates can be exported and processed through mercapturic acid pathways. Glutathione also contributes to protein thiol homeostasis and to recycling of other antioxidants such as ascorbate. Its precise roles vary by tissue, and many regulatory effects observed in laboratory systems remain difficult to quantify in whole organisms.
For solid glutathione, storage conditions affect shelf life. The reduced form is typically kept cool, dry, and protected from air and light. Moisture can promote oxidation, while elevated temperatures accelerate degradation. Suppliers often specify storage at or below freezing, sometimes under inert gas. Solutions are less stable than powders and may require preparation shortly before use. Buffers and chelating agents can slow oxidation, but they do not eliminate it. Published stability data vary with matrix, pH, and container.
Quality control for glutathione focuses on identity, purity, and oxidation state. Certificates of analysis may report assay value, water content, and the presence of GSSG or other impurities. Chromatographic purity is often expressed as a percentage of peak area. Reference standards help laboratories compare results across instruments and batches. Because glutathione is a small, polar molecule, separation from cysteine, gamma-glutamylcysteine, and related thiols can be challenging. Verification often combines more than one analytical technique.
=== Food quality === The level of reducing sugars in wine, juice, and sugarcane are indicative of the quality of these food products, and monitoring the levels of reducing sugars during food production has improved market quality. The conventional method for doing so is the Lane-Eynon method, which involves titrating the reducing sugar with copper(II) in Fehling's solution in the presence of methylene blue, a common redox indicator. However, it is inaccurate, expensive, and sensitive to impurities.
=== Classification (different types of congenital muscular dystrophies) === The subtypes of congenital muscular dystrophy have been established through variations in multiple genes. Phenotype, as well as, genotype classifications are used to establish the subtypes, in some literature. One finds that congenital muscular dystrophies can be either autosomal dominant or autosomal recessive in terms of the inheritance pattern, though the latter is much more common Individuals with congenital muscular dystrophy fall into one of the following types:
Critics accused Mahathir of authoritarianism for centralising power and suppressing dissent, while supporters argued that his actions were necessary to preserve national stability. Mahathir resigned in 2003 after 22 years in office, but remained politically influential and was critical of his successors. He quit UMNO over the 1MDB corruption scandal in 2016, joining BERSATU and leading the Pakatan Harapan opposition coalition to victory in the 2018 general election. During a second tenure as prime minister, he pledged to investigate the 1MDB scandal, combat corruption, and cut spending on large infrastructure projects. He also secured the pardon and release of Anwar Ibrahim. Mahathir resigned in 2020 amidst a political crisis. Despite losing his parliamentary seat in the 2022 general election, he remained active in politics and shifted party affiliation several times. In 2019, Time magazine listed him as one of the world's 100 most influential people. Mahathir's political views have shifted during his life, and are shaped by his Malay nationalism and Islamic religious beliefs.
=== January === 1 January – In his New Year address, Archbishop of Canterbury Justin Welby urges politicians to treat their opponents as human beings rather than enemies. Secretary of State for Defence Grant Shapps says that British forces are ready to act against Houthi rebels targeting cargo ships in the Red Sea. Writing on X, Doug Barrowman, husband of Michelle, Baroness Mone, said it "suits the agenda" of ministers to "scapegoat" him and his wife as a means of distracting from government "incompetence" at failing to procure personal protective equipment during the COVID-19 pandemic. 2 January – The Home Office says it has fulfilled a pledge to clear a "legacy" backlog of 92,000 asylum applications lodged before July 2022. But after it subsequently emerges that over 4,000 cases are still waiting for a decision, the Office for Statistics Regulation (OSR) announces an examination of the figures the next day. 3 January – Sir Ed Davey, leader of the Liberal Democrats, launches the party's election campaign by targeting constituencies with Conservative MPs. Former chancellor of the exchequer Nadhim Zahawi makes a guest appearance in the ITV drama Mr Bates vs The Post Office as himself, questioning Post Office chief executive Paula Vennells in a 2015 House of Commons committee inquiry into the Horizon computer system. Richard Tice, leader of Reform UK, rules out a pact with the Conservative Party at the next general election. 4 January – Prime Minister Rishi Sunak says his "working assumption" is that the general election will take place in the second half of this year.

==== Emulsions ==== Emulsions are mixtures of immiscible liquids. Water-in-oil emulsions of estradiol benzoate were evaluated as long-acting preparations for use by intramuscular injection in the 1940s and 1950s. Formulations of estradiol benzoate alone under the brand name Menformon-Emulsion and with progesterone under the brand name Di-Pro-Emulsion were previously marketed. A 10 mg dose of estradiol benzoate in emulsion by intramuscular injection is said to have a duration of about 2 to 3 weeks. This is similar to the duration of an aqueous suspension of 10 mg estradiol benzoate or an oil solution of 10 mg estradiol valerate. Emulsions of steroids by intramuscular injection have similar properties (e.g., duration) relative to aqueous suspensions. Painful injection site reactions have been reported with emulsions similarly to suspensions.
==== 17OHP subpathway ==== The first step of this pathway is the 5α-reduction of 17OHP to 5α-pregnan-17α-ol-3,20-dione (referred to as 17OHDHP or 17α-hydroxy-dihydroprogesterone). The reaction is catalyzed by SRD5A1. 17OHDHP is then converted to 5α-pregnane-3α,17α-diol-20-one (5α-Pdiol) via 3α-reduction by a 3α-hydroxysteroid dehydrogenase isozyme (AKR1C2 and AKR1C4) or HSD17B6, that also has 3α-reduction activity. The pathway then proceeds from 5α-Pdiol the same way as the pathway that starts from P4, i.e. 5α-Pdiol → AST → 3α-diol → DHT. The pathway can be summarized as: 17OHP → 17OHDHP → 5α-Pdiol → AST → 3α-diol → DHT.
One key to compliance with many of these standards is audit logging of all changes to LIMS data, and in some cases a full electronic signature system is required for rigorous tracking of field-level changes to LIMS data.

== History == The effectiveness and safety of plasminogen is primarily based on one single-arm, open-label (unblinded) clinical trial enrolling 15 adult and pediatric participants with plasminogen deficiency type 1. All participants received plasminogen administered every two to four days for 48 weeks. The effectiveness of plasminogen was demonstrated by at least 50% improvement of their lesions in all 11 participants who had lesions at baseline, and absence of recurrent or new lesions in any of the 15 participants through the 48 weeks of treatment. The U.S. Food and Drug Administration (FDA) granted the application for plasminogen orphan drug designation, fast track designation, priority review, and a rare pediatric disease priority review voucher. The FDA granted approval of Ryplazim to ProMetic Biotherapeutics Inc.
==== Environmental Mitigation ==== Reducing pharmaceutical waste in water systems and soil through improved waste management. Addressing resistance genes in wastewater from hospitals, farms, and drug manufacturing sites.
Additional conservation efforts for the tree involve fencing against livestock, watering of seedlings in open areas, and involving local communities in planting seedlings. By the 21st century threats arising from global warming and overgrazing have made it difficult for new trees to grow in wild. A collaboration of local people with assistance from others such as UK-based Friends of Socotra and Mendel University in Brno has been aiding a number of slow growing saplings by watering them while they are too young to draw down sufficient water, and protecting them from hungry goats. As of 2022, about 600 saplings have reached the point where they no longer need regular watering, leading to hopes that a new generation of the tree may become established on Socotra.
